Research Interests

Our group is interested in developing and applying new chemistry tools to investigate protein-based interactions and activities in living systems. We have been leading the development of bioorthogonal cleavage reactions, which has enabled the in situ gain-of-function study of proteins as well as other biomolecules in living cells and animals. This has opened an exciting new avenue in bioorthogonal chemistry with broad utilities in life sciences and medicine. We have also developed a series of genetically encoded protein labeling, crosslinking and proximity-tagging tools for monitoring biomolecule interactions as well as cell communications during various physiological and pathological processes. Most recently, we started to leverage this exciting toolkit to better understand the mode-of-action of immunotherapy.
